While we previously used the linear interaction\u00a0energy approach to probe the effect of point mutations at protein-protein interfaces, the theoretically exact\u00a0machinery of free energy perturbation represents a better and more accurate alternative. Based on\u00a0comparative bioinformatics analysis of sequences from organisms adapted to different working temperatures key mutations will be identified.\u00a0The temperature dependence characteristic for the mutations will subsequently be examined by performing calculations\u00a0at different temperatures to yield the enthalpy and entropy of the mutations.<\/p>\n<p>Using systems for which experimental data is available will serve to assess and validate the performance of\u00a0the methods and protocols. <a href=\"https:\/\/uit.no\/prosjekter\/prosjekt?p_document_id=228973\">The Norwegian\u00a0Structural Biology Centre<\/a> and <a href=\"https:\/\/arcticzymes.com\">ArcticZymes<\/a> will serve as our experimental collaborations to construct new\u00a0mutants and provide detailed characterizations of their kinetic and thermodynamic properties.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>WP2 will develop the necessary tools to estimate the effect of specific point mutations on the free energy.\u00a0Alchemical transformation of one amino acid into another will be accomplished using the free energy\u00a0perturbation approach combined with soft-core potentials.
