The NMR facility has just now been upgraded with a completely new Bruker system, a 16.4 Tesla Bruker Neo system equipped with an inverse HCN cryogenically enhanced probe (TCI, enhanced for 1H, 13C and 2H), allowing us to fully characterize small amounts of pure unknown material, currently down to approximately 200 µg for an average size small molecule, and the detection limits stretches below 20 µg. For labelled protein samples, a soluble concentration of roughly 0.5 mM is sufficient for a full characterization.
The second system is a Bruker Avance III HD spectrometer operating at 400 MHz for protons, equipped with a broadband SmartProbe and a sample case. The instrument is open access and operates in full automation.
NMR is an extremely versatile method based on the behavior and interaction between magnetic spins of atomic nuclei. Different sets of experiments provide information about the bonding network, the 3D configuration and conformation in solution, as well as information about dynamics on different time scales.
